Gloria is a hardworking, though very stressed, café barista. She gets irritated with the Greens when they mess up her café. Already quite stressed due to the load of her job, Gloria can quickly lose her patience, especially when it has something to do with Cricket's shenanigans. However, she can stoop to Cricket and Tilly's level of childishness such as when she stole their critterballs out of spite and eventually started playing with them before almost getting fired by her boss ("Critterball Crisis"). Once Cricket begins to work at Big Coffee, she is still somewhat annoyed by his antics, but has cooled down and has developed something of an older sister attitude to the boy. Gloria dreams of making enough money so that she may see Paris, which she has a romantic view of to the point of Paris Syndrome and is why she doesn't want Cricket to jeopardize her dreams. She has an artistic side, as revealed in "Paint Misbehavin'", and has dabbled unsuccessfully in abstract art.

Barry is a sweet, sensitive, and mellow bunny. In contrast to the energetic and adventurous Kiff, Barry is more composed and timid in nature, which means that he can sometimes find himself hesitant to proceed with the situations that Kiff drags him into. At the same time, these qualities also allow him to act as a voice of reason whenever Kiff gets in over her head.

Kiff is a generally cheerful, optimistic, and helpful squirrel whose ambitious and adventurous nature leads her into many escapades. During these adventures, she often tries to spread positivity and help people to the best of her abilities. She is a highly determined individual, a trait which typically manifests itself in her adventures, sometimes to a fault; her determination can occasionally lead her into unnecessarily complicated situations. Kiff also has a great deal of love for her family and friends, and is willing to stick up for them at all costs. Nonetheless, Kiff can also be highly cunning if the situation calls for it. While generally respectful of authority figures and rules, Kiff is not above defying them should they get in the way of her personal principles or goals, or even her curiosity. In particularly hectic situations, she has been known to resort to what she calls "squirrel attacks", which typically involve her aggressively leaping onto others.

Ami is known for her girly-girl and goody-two-shoes personality. As the optimist, Ami always looks on the bright side of things and finds a solution to every situation she is involved in. She loves rainbows, bunnies (cute animals), sparkles and anything else typically pretty, which is the opposite of Yumi, who likes things more targeted to a tomboyish and unisex demographic. She also cares a lot about how other people think, and tries not to be selfish. This makes Ami willing to help everyone and find solutions for anyone she cares. On the other side, however, she also hides her real mind and opinions in order to never disappoint others. In the pilot, she was depicted being less comical.

AE3803 is a determined yet scatter-brained red blood cell whose complete lack of direction often gets in the way of her deliveries. She is also Eli Ketchum's mother.
