In They Stole Max's Brain, Sam shows more violent manner of investigating than the previous episodes, which include from beating a gorilla for showing a dog sign to using a gun on a COPS member. It is usually, and somewhat ironically, Max that calms him down and prevents him from acting upon his anger. Although most of the time he acts like a regular human, Sam occasionally behaves like a dog, having an enhanced smell and often making several dog sounds, such as growling or whining. Max is Sam's long-time best friend and sidekick. They met as children and attended junior high together before becoming the Freelance Police. In most situations, Sam plays the straight man against Max's chaotic behavior. Sam is incredibly close to Max and will become tangibly uncomfortable when separated for any reason; he is willing to delve into great lengths to protect his little buddy. Unlike most people, Sam seems to admire Max's violent tendencies, for the most part.

Michelangelo is a darkish verdant green skinned terrestrial chelonian mutant/yokai who possesses the features of an ornate box turtle, sporting an altered human-like physique consisting of sculpted, yet overall petite figure. Being the shortest among his brothers (both probably due to him being the youngest as well as his miniature turtle species), having three fingers on each hand, and two toes on each foot. His turtle features consist of a bright yellow plastron, a blue grey carapace that has a somewhat darker outer trim around the edges, followed with a bright sunflower yellow pattern displayed on the back. The pattern resembling very closely to the curly bracket symbol {}; Finally, on each side of his shoulder blades, alongside his upper thighs, are matching yellow spots that have a faint orange outline surrounding them. Mikey's eyes are onyx and rather doe, meant to accentuate his role of being the youngest brother. Mikey is playful and lighthearted. He is intuitive, able to adopt new skills quickly, determine others' problems, and at times accurately predict situational outcomes. Whenever he gets scared or feels scared, he retreats his whole body into his shell. He is an artist and spends his time tagging the Turtles' lair and his shell with graffiti. Mikey has three personas he created: Doctor Delicate Touch, Doctor Feelings, and Doctor Positive. To deliver a harsh truth in his "Doctor Delicate Touch" method, Mikey often starts off being sweet, but quickly becomes angry and frank. Doctor Feelings uses his emotional intelligence to advise others, and Mikey faces issues with determined optimism as Doctor Positive. He owns a pair of nunchaku that he can extend nigh-infinitely to grapple things and opponents, using his mystical powers bestowed in him from the ancestors of the Hamato clan.

James is based on a Lancashire and Yorkshire Railway (L&YR) Class 28, a class of 0-6-0 mixed-traffic tender locomotive designed by George Hughes for the Lancashire and Yorkshire Railway and built by Horwich Works between 1909 and 1912. James is very proud of his splendid red paintwork and likes to let everyone know it. A large downside to James' vanity is that it makes him prone to acts of selfish entitlement and being rude, inconsiderate and overly stubborn until he gets his way, from picking on other smaller engines and trying to take another engine's duties to making false accusations and refusing to take accountability for an accident.
