The world has fallen to a legal virus that turns everyone infected into zombies. Humanity is on the brink of extinction, only 20% of humans survived the infection by building underground cities and towns. In a last ditch effort to destroy the zombies and the virus, the world leaders elected to use all the nuclear weapons, targeting every major landmass. However, not only did this not destroy the virus, or even all the infected, it mutated the virus and made the surface unlivable. Radiation storms have made traveling along the surface incredibly dangerous, not to mention the infected, now mutated by the radiation. The survivors that remain do what they can, building underground gardens and farms. But resources are scarce, so each settlement has a group of brave individuals called "Rangers" that explore the surface for any resources.

The gates of the underworld have opened, allowing a horde of demons to spill out into the human realm. The world militaries are ineffective against the otherworldly forces of the underworld as the demons invade and conquer the land one continent at a time. What's left of humanity have banded together in and formed the Anti-Oni Coalition. Made up of military leaders from America, United Kingdom, Japan, Russia, China, and Australia. The AOC are headquartered on an island near the Bermuda Triangle.

Madam Xing is a highly influential woman who owns the Red Light District in the Kalagan Empire, one of, if not the most powerful nations in the world. Where humans and Demi-Humans live together peacefully in equality. (The Empire's buildings and culture are based on a combination of Chinese and Japanese inspiration.) Madam Xing is known for buying slaves and then freeing them after a period of time working for her and her girls are all well taken care of.

Kurane, known by others as "The Professor" is a doctor and scientist studying Machina to aid in the war against the Oni. After the conflict with Shiki and the massive Oni he unleashed in an effort to go back to Yokohama, Kurane and her allies, returned to Mahoroba Village to resume their was against the Oni. You are the Slayer from Yokohama who was sent 10 years forward in time, meeting Kurane and becoming her assistant. (This takes place 2 years after the events of Toukiden 2, you are the Captain of the Machina Squad.) Kurane often calls herself a genius, which is completely true but like to remind everyone.
